470 Infantile hemangioma telocytes and noradrenaline are the prerequisite for response to propranolol

چکیده
Propranolol, a nonselective β-adrenergic receptor blocker, is the first-line medical treatment for severe infantile hemangiomas (IH). We have shown in mice that propranolol-induced tumor growth inhibition was correlated with altered expression of Aquaporin-1 (AQP1), an aqueous channel modulated angiogenesis and cell migration. In IH, AQP1 made it possible to identify interstitial named telocytes (TC). TC are characterized by long cytoplasmic processes, telopods, which form three-dimensional communication network between cells, organs such as skin, particular dermal vessels. The localization marker IH compared other hemangiomas. Defining new pool types interest we isolated, cultured studied lesional: endothelial cells (EC), pericytes (PER) TC. Using vitro model developed, response propranolol function shRNA. Then, since betablockers antagonize effect natural ADRB agonists, postulated catecholamines produced situ may role response. Quantifying tissues, found high amount noradrenaline (NA) untreated proliferative involuted or propranolol-treated IH. further first three enzymes catecholamine biosynthesis pathway expressed their levels reduced tumors. performed assay IH-EC, PER were incorporated. Results showed total number tubes formed sensitive only when NA added 3-cell model. conclude sensitivity depends on crosstalk lesional EC, AQP1+ context local NA.

Propranolol therapy for infantile hemangioma.
CONTEXT There has been widespread interest surrounding the use of beta-blockers (i.e. propranolol, timolol, nadolol, acebutolol) in the treatment of infantile hemangiomas (IH). OBJECTIVE To review literature evaluating treatment of IH with propranolol. EVIDENCE ACQUISITION We conducted a literature search on PubMed and investigated for case reports, case series, and controlled trials by usi...
متن کاملWhen to stop propranolol for infantile hemangioma
There is no definitive conclusion regarding the optimal timing for terminating propranolol treatment for infantile hemangioma (IH). A total of 149 patients who underwent detailed color Doppler ultrasound examination were included in this study. The characteristics and propranolol treatment of all patients were summarized and analyzed. Patients were divided into two groups according to the lesio...
متن کاملExcellent response of infantile orbital hemangioma to propranolol
Infantile hemangiomas are the most common vascular neoplasm that present in infancy, with more than half affecting the head and neck region. Periocularly, hemangiomas may be complicated by visual loss through induction of strabismal, deprivational, or anisometropic astigmatism. We report a case of a 5-year-old girl who presented with orbital hemangioma with potential risk of visual loss who had...
متن کاملPropranolol therapy for infantile hemangioma: our experience
OBJECTIVE Hemangiomas are the most common benign vascular tumors of infancy. Although most infantile hemangiomas (IHs) have the ability to involute spontaneously after initial proliferation and resolve without consequence, intervention is required in a subset of IHs, which develop complications resulting in ulceration, bleeding, or aesthetic deformity. The primary treatment for this subset of I...
متن کاملMechanisms of propranolol action in infantile hemangioma
Infantile hemangioma is a common tumor of infancy. Although most hemangiomas spontaneously regress, treatment is indicated based on complications, risk to organ development and function, and disfigurement. The serendipitous discovery of propranolol, a non-selective β-adrenergic receptor blocker, as an effective means to regress hemangiomas has made this a first-line therapy for hemangioma patie...
